Chemical cartography of host-pathogen-microbiome interactions

Spatial context is essential to understand host-microbe-environment interactions and link chemical structure to biological function. Using an approach called “chemical cartography”, we combine liquid chromatography tandem mass spectrometry (LC-MS/MS), 3D modeling and data analytics with infection biology, to identify the spatial distribution of molecules and how they relate to infectious disease pathogenesis and microbiota dysbiosis. In this presentation, I will illustrate the power of this approach as applied to Trypanosoma cruzi infection, causative agent of Chagas disease. Chagas disease symptoms include cardiac arrhythmias, cardiac apical aneurysms, enlargement of the oesophagus and of the colon (megaoesophagus and megacolon). Our approach revealed persistent metabolic alterations at the sites of Chagas disease symptoms in our infection model. Furthermore, it led to the discovery of a new candidate treatment for Chagas disease and a new, metabolism-based perspective on Chagas disease tolerance. Current work is demonstrating the broad applicability of this approach across infectious diseases.
